In vivo CAR-T therapy is reshaping how cell therapies could be developed and delivered, shifting from complex, multi-step manufacturing toward a streamlined, in-body approach. Lipid nanoparticle (LNP) systems are emerging as a key enabler, allowing genetic material to reach and reprogram T cells directly within the patient. Advances in LNP design are improving delivery beyond the liver, with promising targeting of lymphoid organs like the spleen, where immune activity is concentrated. Enhanced specificity across splenic cell types, including T cells, points to new possibilities for efficient, scalable immunotherapy. By removing the need for cell extraction and reinfusion, this approach could reduce timelines and broaden patient access.
