Expand mRNA-LNP Nanomedicine Potential By Optimizing Lipids

Getting nucleic acids to the right cells intact is still one of the hardest problems in gene and cell therapy development. This article looks at how small changes to lipid nanoparticle composition can alter delivery efficiency, including a fifth component that redirects delivery beyond the liver. It sorts ionizable lipids into five structural categories, each with its own trade-offs in potency, toxicity, and clinical translation. Commentary from Dr. Anna Blakney, Assistant Professor at the Michael Smith Laboratories and School of Biomedical Engineering at UBC, describes formulation as both an art and a science. A featured iPSC-derived neuron study shows why a formulation that encapsulates better is not automatically the better choice.
