Development And Optimization Of LNP Formulations Using The IJM NanoScaler

The demand for robust, scalable, and reproducible Lipid Nanoparticle (LNP) formulations for nucleic acid delivery is on the rise. Curapath and KNAUER joined forces to investigate the potential of KNAUER's Impingement Jets Mixing (IJM) NanoScaler in optimizing LNP formulations for maximized encapsulation efficiency and reproducibility.
The study assessed the impact of key operational parameters, including Total Flow Rate (TFR), on the size, Polydispersity Index (PdI), and encapsulation efficiency of LNPs. Results indicated that the NanoScaler matched the performance of traditional Herringbone mixers in producing LNPs with comparable characteristics. Notably, the NanoScaler offered enhanced control and flexibility, allowing for size tuning by TFR adjustments and in-line dilution to minimize ethanol concentration without compromising LNP properties. The study further demonstrated successful scalability from R&D to larger batches for preclinical studies using different IJM mixers.
