T7 Compatibility Is Not The Same As T7 Performance

T7-based IVT workflows remain the default across RNA development, largely because of familiarity: existing templates, established protocols and CDMO processes built around them. That familiarity, however, can obscure meaningful differences in yield, dsRNA formation, capping flexibility and translatability between workflows that are all technically "T7-compatible."
This article reframes the evaluation question many RNA teams skip. Rather than asking whether a workflow fits existing T7-promoter templates, it asks whether that workflow can improve synthesis quality and reduce downstream burden without forcing unnecessary redesign. It outlines where upstream IVT choices, not just downstream purification, shape impurity profiles, and offers a set of specific questions teams can use to distinguish genuine process readiness from simple familiarity.
