ExTend Cap AU is an RNA polymerase designed to maximize yield and capping efficiency during IVT of self-amplifying or self-replicating mRNA.

What level of capping efficiency can I expect to see from co-transcriptional capping with an AU cap analog?

Achieve >95% capping efficiency with as little as 1 mM cap, lowering reagent requirements.

What type of yield can I expect for my self-amplifying RNA?

ExTend Cap AU delivers consistently higher yield than T7 while maintaining integrity.

What about dsRNA, how does this enzyme perform?

All Prima RNApols minimize dsRNA to almost undetectable levels, reducing downstream purification needs and improving the safety and performance of mRNA therapeutics.

What other benefits can I expect from this enzyme?

Cost-efficient manufacturing. Subject to IVT optimization, we’ve shown use of less template (4 nM), enzyme (5 U/µL IVT), and cap analog (1 mM) can provide significant cost savings.

Does ExTend Cap AU work with the T7 promoter?

No, ExTend Cap AU is only compatible with the ExTend Cap AU promoter. The specific promoter sequence must be incorporated into the DNA template of interest to successfully generate mRNA during IVT. For information about this process, refer to the Use Guide or contact Primrose for support.

Will ExTend Cap AU work with any transcription start site?

ExTend Cap AU is an engineered enzyme optimized for co-transcriptional capping with AU cap analogs. However, ExTend Cap AU is also compatible with other cap analogs and transcription start sites.

Can I use my preferred capping strategy?

ExTend Cap AU is optimized for AU co-transcriptional capping, significantly improving efficiency and reducing reagent use for saRNA templates.

What does the kit contain?

The ExTend Cap AU kit contains 20,000 U RNA polymerase, a 9.5 kb linearized saRNA template, and 5x reaction buffer.

How was performance measured?

Capping efficiency is determined by LC-MS. dsRNA is determined by J2 ELISA. RNA integrity is determined using Agilent Fragment Analyzer. Yield is measured using A260.
